At Allele Biotechnology, we have embraced a footprint free method to reprogram adult cells to iPSCs using modified mRNA molecules. We are working meticulously alongside Dr. Luigi Warren to enhance this technology and offer it to our customers as a custom service.

Method of Generation

• mRNA - Non-integrating solution for genetically unmodified iPSCs that more closely resemble hESCs. Xeno free RiPSCs can be generated if desired.

Required Materials

• 1 - 2 vials of each cell type and indication of appropriate growth medium and conditions. Currently, our service is for fibroblast cells, but other cells can be evaluated on a case-by-case basis.

Deliverables

• We typically aim to deliver 5 - 6 lines per starting cell type. You may specify the desired cell count as well as the number of lines.

Duration

• Project times vary based on amount of analysis and desired cell count delivery. A standard mRNA transfection can be completed in about 1 to 2 weeks, and a project with full analysis can take up to 3-6 months.

Other methods of generation available include:

• Lentivirus - lentiviral delivery of 4 reprogramming factors (Oct4, Sox2, klf4, cMyc) using a polycistronic lentivirus.

• Retrovirus - Retroviral delivery of 4 reprogramming factors (Oct4, Sox2, klf4, cMyc).

• Customer specified - In addition to OSKM, we also carry Lin28, Nanog, hTERT, and p53 shRNA. We can design a custom plan utilizing any of these additional factors.
