mScarlet3

mScarlet3 has the record high brightness as a red fluorescent protein, and in cells, more than fivefold brighter than mCherry. With a pKa=4.2, mScarlet3 has strong acid resistance, particularly beneficial for endomembrane studies. On the other end of the pH range, this fluorescent protein retains 50% of fluorescence for over 40 minutes at pH13.5. mScarlet3 displays monoexponentially fluorescence decay with long lifetimes of 4.0 ns, facilitating possible application as donors in FRET with fluorescence lifetime imaging (FLIM).

Based on comparison studies, mScarlet3 is the best FRET acceptor of YFPs. As a matter of fact, it will make an outstanding FRET pair with Allele’s other popular fluorescent protein in the green/yellow spectrum—mNeonGreen!
