mScarlet3 has the record high brightness as a red fluorescent protein, and in cells, more than fivefold brighter than mCherry. With a pKa=4.2, mScarlet3 has strong acid resistance, particularly beneficial for endomembrane studies. On the other end of the pH range, this fluorescent protein retains 50% of fluorescence for over 40 minutes at pH13.5. mScarlet3 displays monoexponentially fluorescence decay with long lifetimes of 4.0 ns, facilitating possible application as donors in FRET with fluorescence lifetime imaging (FLIM).
