Custom iPSC Generation


Allele Biotech has offered iPS solutions since iPSC was first reported in 2006, with a focus on the latest technology to advance and simplify iPS generation of ground-state, truly high quality iPSC lines. We carry all iPS factors on retrovirus, lentivirus, and as mRNA for truly footprint free iPS.While we use optimized cocktails of factors and fusions with unique protocols developed at our own facility, commercial customers need to realize that some of the factors may be under patent protection when used for certain purposes.

Next Gen mRNA Reprogramming

In 2010, Warren et al. reported a novel, footprint-free method to reprogram human fibroblasts into iPSCs using messenger RNA.  In our latest publication, "Feeder-Free Derivation of Human Induced Pluripotent Stem Cells with Messenger RNA" which appears in Scientific Reports, Dr. Warren and the team of scientists at Allele Bitoechnology present a substantially enhanced mRNA reprogramming protocol.  By harnessing engineered reprograming factors and optimizing various experimental conditions we can now offer a feeder-free, xeno-free protocol that can reprogram human skin cells to pluripotency in under two weeks.

As the developers of this new iPSC generation method, scientists at Allele offer customers a highly advanced reprogramming technology for a multitude of different cell types. Reprogrammed cells will be completely footprint-print free, characterized, validated, and ready to proceed to a variety of downstream applications. The challenges and ensuing troubleshooting commonly associated with iPS reprogramming are comfortably handled by experienced scientists who have performed numerous reprogramming related experiments. If required, Allele can also reprogram desired cells under GMP procedure for potential therapeutic uses. 

Custom Service details

Required Materials: Customer-desired cells, 1 vial of typical storage size, and indication of appropriate growth medium and conditions. Currently most iPSC lines were created from fibroblast-type of primary cells; other cell types may not be appropriate using the current protocols.

Deliverables:  We typically aim to deliver lines from 3 colonies (sometimes some lines may die off in the process) Customer specified quantity of cells or number of lines in customer specified format can be discussed.

Duration: 4 - 12 weeks generally, actual time will vary based on method chosen and additional customer requirements.
